用放射免疫法和高效液相色谱电化学检测器分别测定了SAMP8小鼠血浆、大脑皮层、海马、下丘脑皮质酮(corticosterone,CORT)水平和大脑皮层、海马兴奋性氨基酸含量。结果显示,SAMP8小鼠血浆CORT水平均显著(P<0.05,或P<0.01)高于其同龄正常对照小鼠SAMR1。SAMP8小鼠8月和12月龄大脑皮层和海马CORT水平也明显(P<0.05,或P<0.01)高于同龄正常对照SAMR1小鼠。同时,SAMP8小鼠大脑皮层和海马谷氨酸(glu-tamate,Glu)含量有上升趋势,谷氨酰胺(Glutamine,Gln)含量则明显高于SAMR1小鼠。SAMP8小鼠切除肾上腺后,大脑皮层和海马Glu含量呈下降趋势,而Gln含量则显著降低。给切除肾上腺的SAMP8小鼠补充CORT(ip,10mg/kg),大脑皮层和海马Glu和Gln水平均明显(P<0.05,或P<0.01)升高,而天冬氨酸(Aspartate,Asp)含量始终没有变化。结果提示,SAMP8小鼠大脑皮层和海马Glu和Gln水平升高是肾上腺依赖的。SAMP8小鼠大脑皮层和海马Glu和Gln含量升高可能与其大脑皮层和海马CORT水平升高有?
The plasma, cortex, hippocampus, hypothalamic corticosterone (CORT) levels and excitatory amino acids in the hippocampus of SAMP8 mice were measured by radioimmunoassay and high performance liquid chromatography-electrochemical detector respectively. The results showed that plasma CORT levels in SAMP8 mice were significantly (P <0.05 or P <0.01) higher than their normal SAMR1 mice of the same age. CORT levels in cerebral cortex and hippocampus of SAMP8 mice at August and 12 months were also significantly higher (P <0.05 or P <0.01) than SAMR1 mice of the same age. At the same time, the content of glutamate (Glu-tamate, Glu) in cerebral cortex and hippocampus in SAMP8 mice increased, and the content of glutamine (Gln) was significantly higher than that in SAMR1 mice. After excision of the adrenal glands in SAMP8 mice, Glu levels in the cerebral cortex and hippocampus decreased, whereas Gln levels decreased significantly. Addition of CORT (ip, 10 mg / kg) to adrenalectomized SAMP8 mice resulted in a significant (P <0.05 or P <0.01) increase in both Glu and Gln levels in the cerebral cortex and hippocampus, while aspartate ( Aspartate, Asp) content has not changed. The results suggest that elevated levels of Glu and Gln in the cerebral cortex and hippocampus of SAMP8 mice are adrenal-dependent.
