目的:探讨人野生型Rb基因转移对预防血管成形术后再狭窄的作用。方法:通过逆转录病毒载体介导,将野生型Rb基因转导至大鼠球囊损伤的颈动脉再狭窄模型中,21天后观察其对新生内膜形成的影响,并检测Rb蛋白表达水平。结果:成功地构建和包装了人野生型Rb基因的重组逆转录病毒载体,并在体外细胞中成功表达;免疫组化检测表明转导血管局部表达人野生型Rb蛋白,与对照组相比,损伤血管新生内膜/中层面积比降低了49%。结论:人野生型Rb基因治疗时血管成形术后再狭窄有一定的预防作用。
Objective: To investigate the role of wild-type Rb gene transfer in preventing restenosis after angioplasty. METHODS: The wild-type Rb gene was transduced into the model of carotid artery stenosis induced by balloon injury in rats by retroviral vector. The expression of Rb protein was detected after 21 days. Results: The recombinant retroviral vector containing human Rb gene was successfully constructed and packaged and successfully expressed in vitro. Immunohistochemistry showed that the wild-type Rb protein was expressed locally in transduced blood vessels. Compared with the control group, Intimal injury / neointimal area ratio decreased by 49%. Conclusion: The restenosis after human angioplasty has some preventive effects on human wild-type Rb gene therapy.
