方法:用氯化钾诱发SD大鼠皮层扩散性抑制(CSD),改良 Longa法制作大鼠 MCAO模型,用地高辛标记的 cRNA探针,行原位杂交检测谷氨酸载体EAAC1 mRNA表达。结果:假手术组脑片上单位面积OD为(2 189±263),阳性颗粒数为(43.2±4.7)个。缺血 30min时皮层EAAC1 ,mRNA表达明显上调,经CSD处理后再缺血时,与对照组比较表达水平均明显增高,其中Li 30min时最高。结论:CSD的作用可能与诱导EAAC1合成,增加谷氨酸的摄取有关。
Methods: CSD was induced by potassium chloride (CSD). MCAO model was established by modified Longa method. The glutamate transporter EAAC1 mRNA was detected by in situ hybridization with digoxigenin-labeled cRNA probe. Results: In the sham operation group, the unit area OD of the brain slices was (2 189 ± 263) and the number of positive particles was (43.2 ± 4.7). The expression of EAAC1 and mRNA in the cortex was significantly increased at 30min after ischemia, and the expression of EAAC1 and mRNA was significantly increased in the ischemic group after CSD treatment. Conclusion: The effect of CSD may be related to induction of EAAC1 synthesis and increase of glutamate uptake.
