目的观察慢性高原病(CMS)患者骨髓CD71~+、CD235a~+有核红细胞增殖、凋亡变化。方法提取16例CMS患者(CMS组)及16例单纯陈旧性骨折患者(对照组)骨髓单个核细胞(BMMNCs),用免疫磁珠法分选CD71~+、CD235a~+有核红细胞。用CCK-8法检测骨髓CD71~+、CD235a~+有核红细胞增殖能力,Annexin V-FITC/PI双标记流式细胞术测算细胞凋亡率,集落形成实验测算骨髓CD71~+、CD235a~+有核红细胞及BMMNCs克隆形成率。结果与对照组比较,CMS组骨髓CD71~+、CD235a~+有核红细胞增殖能力升高,凋亡率降低(P均<0.05)。培养第7、14天,两组骨髓CD71~+、CD235a~+有核红细胞克隆形成率比较,P均>0.05;两组BMMNCs克隆形成率比较,P均<0.05。结论 CMS患者骨髓CD71~+、CD235a~+有核红细胞增殖增加、凋亡减少。
Objective To observe the changes of proliferation and apoptosis of CD71 +, CD235a ~ + erythroblasts in patients with chronic mountain sickness (CMS). Methods Bone marrow mononuclear cells (BMMNCs) from 16 patients with CMS (CMS group) and 16 patients with simple fracture (control group) were collected and sorted into CD71 ~ + and CD235a ~ + nucleated erythrocytes by immunomagnetic beads method. The proliferative ability of CD71 +, CD235a ~ + erythroblasts was detected by CCK-8 method. The apoptotic rate was measured by Annexin V-FITC / PI double labeling flow cytometry. Nucleated red blood cells and BMMNCs clone formation rate. Results Compared with the control group, the proliferative ability of CD71 +, CD235a ~ + nucleated erythrocytes in CMS group was decreased and the apoptosis rate was decreased (all P <0.05). On the 7th and 14th day of culture, the colony formation rates of CD71 ~ +, CD235a ~ + nucleated erythrocytes in two groups were all higher than that of the control group (P> 0.05). The colony formation rates of BMMNCs in both groups were all lower than 0.05. Conclusions The proliferation of CD71 +, CD235a ~ + nucleated erythrocytes in CMS patients is increased and the apoptosis is decreased.
