目的 通过体外实验研究 ,探讨胰岛素、上皮生长因子 (EGF)对人体消化系统 4种肿瘤细胞株 ,即人胃腺癌细胞株 30 5 1 80、人胰腺癌细胞株PaTu 890 2、人结肠腺癌细胞株SW 480和HT 2 9增殖生长的影响。方法 配置各细胞株生长所需的介质液 ,其中加入不同浓度胰岛素 (0 2 ,2 ,2 0 ,2 0 0 ,2 0 0 0mu/ml)或胰岛素 +EGF(1ng/ml) ,置各细胞株的一定数量细胞于相应介质液中培养 2 4h ,用 3H Thymidine掺入法检测细胞增殖状况。 结果 30 5 1 80在胰岛素或胰岛素 +EGF的介质液中迅速增殖 (P <0 0 5 ) ,且与胰岛素呈浓度依赖关系 ;PaTu 890 2在胰岛素浓度为 0 2 ,2 ,2 0mu/ml时增殖明显 ,在同样胰岛素浓度 +EGF的介质液中也表现出相似的效应 (P <0 0 5和P <0 0 1) ,但在其它浓度时无显著变化 ,SW 480的增殖仅在胰岛素浓度为 2 ,2 0 ,2 0 0mu/ml时明显 (P <0 0 5 ) ;而对HT 2 9细胞株 ,胰岛素或胰岛素 +EGF对其增殖的刺激作用均不大 (P <0 0 5 )。在本实验中 ,未见到胰岛素和EGF对细胞增殖的明显协同效应。结论 人体消化系统的肿瘤对胰岛素和EGF促细胞增殖作用可因其细胞株的不同反应各异。
Objective To investigate the effects of insulin and epithelial growth factor (EGF) on the human digestive system in 4 tumor cell lines: human gastric cancer cell line 30 5 180, human pancreatic cancer cell line PaTu 890 2, and human colon adenocarcinoma cell. Effects of Proliferation and Growth of SW 480 and HT 2 9 Strains Methods The medium required for the growth of each cell line was configured, and various concentrations of insulin (02, 2, 20, 200, 200 mu/ml) or insulin + EGF (1 ng/ml) were added to set the cells. A certain number of cells in the strain was cultured in the corresponding medium for 24 hours, and cell proliferation was detected by 3H Thymidine incorporation method. RESULTS: The rapid proliferation of 3051 80 in insulin or insulin + EGF medium (P < 0.05) and insulin-dependent concentration; PaTu 890 2 at an insulin concentration of 0 2, 2, 20 mu / ml Proliferation was significant, and similar effects were also observed in the media with the same insulin concentration and EGF (P < 0.05 and P <0 01), but no significant changes at other concentrations. The proliferation of SW 480 was only at the insulin concentration. It was significant at 2, 20, 200mu/ml (P < 0.05), whereas the effect of insulin or insulin + EGF on the proliferation of HT29 cells was not significant (P <0 05). . In this experiment, no significant synergistic effect of insulin and EGF on cell proliferation was seen. Conclusions Tumors of the human digestive system have different effects on cell proliferation due to insulin and EGF promoting cell proliferation.
