Investigating the Release of Myristoylated Ciliary Proteins Using Synthetic Peptides

  Lipid modification which includes S-prenylation,S-palmitoylation and N-myristoylation is one of the common post-translational modifications of proteins.Lipidated proteins play an important role in cellular activity and cell signaling.N-myristoylated proteins are involved in many important signaling pathways and related with some diseases.Although the delivery of prenylated cargo has been identified,the release of the myristoylated cargo from its solubilizing factors UNC119 is not well understood.We synthesized the myristoylated ciliary peptides and further explored their release from UNC119 under the regulation of Arl3 using fluorescence polarization assay and structural analysis.We conclude that ciliary distribution of myristoylated proteins depends on both nucleotide status and cellular localization Arl3.
