人红细胞游离嘌呤核苷酸水平是估价非胰岛素依赖型糖尿病NIDDM疾病过程的重要参数。本文采用反相高效液相谱法(reversed-phase high performance liguid chromatography,rHPLC)测定NIDDM患者和健康人红细胞胞液中ATP、ADP、AMP和GTP的含量。红细胞经10%HClO_4处理,离心后收集上清液,加入K_2CO_3中和后离心取上清液进样。本法灵敏度可靠、特异性高。实验发现:NIDDM病人的ATP/AMP比正常人低65%,而HbA_1c为正常人的两倍,ATP含量低和HbA_1c升高呈正相关(r=0.92)。因此,测定红细胞中的腺苷酸和HbA_1c含量是研究糖尿病人糖类代谢和能量代谢紊乱的有效方法。
Human erythrocyte free purine nucleotide levels are important parameters in assessing NIDDM disease progression in non-insulin dependent diabetes mellitus. In this paper, reversed phase high performance liquid chromatography (rHPLC) was used to determine ATP, ADP, AMP and GTP levels in NIDDM patients and healthy human erythrocyte cytosol. Erythrocytes were treated with 10% HClO_4, the supernatant was collected after centrifugation, centrifuged by adding K_2CO_3 and the supernatant was injected. This method is sensitive and specific. The results showed that ATP / AMP in NIDDM patients was 65% lower than that in normal subjects, while HbA 1c was twice as high as normal subjects. The low ATP level was positively correlated with the increased HbA 1c (r = 0.92). Therefore, the determination of adenosine and HbA_1c levels in erythrocytes is an effective method to study the disorder of glucose metabolism and energy metabolism in diabetic patients.
