目的建立同时测定不同产地的益智的不同部位中5-羟甲基糠醛(1),teuhetenone A(2),杨芽黄素(3)和诺卡酮(4)含量的超高效液相色谱法(UPLC)。方法采用ACQUITY UPLC,HSS T3色谱柱(2.1 mm×100 mm,1.8μm),流动相为乙腈-0.1%的甲酸水溶液梯度洗脱,流速为0.5 mL·min-1,检测波长采用255 nm。结果化合物1,2,3和4分别在1.223~24.46,2.016~40.32,1.875~37.50和16.78~335.6μg·mL-1线性关系良好,平均回收率分别为99.5%,101.5%,100.9%和101.2%。结论化合物2和4主要分布于种子中,而化合物1和3均分布于种子和果壳中;该方法快速、简单、重复性好,可以为益智的鉴别和质量控制提供依据。
OBJECTIVE To establish a method for the simultaneous determination of 5-hydroxymethylfurfural (1), teuhetenone A (2), pyridium (3) and nootkatone (4) in different parts of different areas by using ultra performance liquid chromatography Act (UPLC). Methods ACQUITY UPLC, HSS T3 column (2.1 mm × 100 mm, 1.8 μm) was used. The mobile phase consisted of a gradient of acetonitrile-0.1% formic acid in water with a flow rate of 0.5 mL · min-1 and a detection wavelength of 255 nm. Results The linear relationships of compounds 1, 2, 3 and 4 were 1.223 ~ 24.46, 2.016 ~ 40.32, 1.875 ~ 37.50 and 16.78 ~ 335.6μg · mL-1 respectively.The average recoveries were 99.5%, 101.5%, 100.9% and 101.2 %. Conclusion Compounds 2 and 4 are mainly distributed in seeds, while compounds 1 and 3 are distributed in seeds and husks. The method is rapid, simple and reproducible, which can provide the basis for identification and quality control of the puzzle.
