观察能量可控陡脉冲对乳腺癌细胞增殖抑制作用以及相关基因表达的影响,探讨能量可控陡脉冲抑制肿瘤细胞增殖的可能机制。能量可控陡脉冲作用于人乳腺癌细胞系MDA-MB-231,于作用后24h应用TUNEL法观察肿瘤细胞凋亡,RT-RCR方法测定处理组和对照组中Rb基因及转录调节因子E2F1基因表达的改变。陡脉冲对乳腺癌细胞的增殖有明显抑制作用,与对照组相比处理组细胞数目明显减少,TUNEL阳性细胞显著增多,处理组Rb基因mRNA表达明显升高、E2F1mRNA表达明显下降。能量可控陡脉冲具有抑制肿瘤细胞增殖,诱导肿瘤细胞凋亡的作用,Rb基因、EF基因可能参与了能量可控陡脉冲抑制恶性肿瘤细胞增殖的过程。
To observe the effect of energy-controlled steep pulses on the proliferation inhibition of breast cancer cells and the expression of related genes, and to explore the possible mechanism that steep pulses can inhibit the proliferation of tumor cells. The energy-controlled steep pulses were applied to the human breast cancer cell line MDA-MB-231. TUNEL method was used to observe the apoptosis of tumor cells 24 h after treatment. RT-RCR method was used to determine the Rb gene and transcriptional regulator E2F1 gene Changes in expression. The steep pulse significantly inhibited the proliferation of breast cancer cells. Compared with the control group, the number of cells in the treated group decreased significantly, the number of TUNEL positive cells increased significantly, and the expression of Rb mRNA and E2F1 mRNA in the treatment group decreased significantly. The energy-controlled steep pulses can inhibit the proliferation of tumor cells and induce the apoptosis of tumor cells. The Rb gene and EF gene may be involved in the energy-controlled steep pulses to inhibit the proliferation of malignant tumor cells.
