为了深入了解芥子气的血液毒理学的特征,观察了骨髓CFUs及CFUc的变化,并与~(60)Co照射比较。芥子气作用下的CFUs剂量-存活曲线为具有早期肩区的指数性直线,其特征是n为5.6,D_0值为13.7mg/kg,而~(40)Co照射的指数性直线其n=1.1,D_0=125rad。在等毒性剂量LD_(50)作用下的芥子气CFUs的存活率比辐射损伤大10倍,说明CFUs虽对芥子气很敏感,但其敏感性低于辐射损伤。用3/4LD_(60)等毒性剂量芥子气,CFUs增殖开始早,于中毒后4~18天恢复正常,而~(60)Co照射后7~36天才恢复到正常的一半。35mg/kg大剂量芥子气中毒后第1天的CFUs或CFUc存活率均低于正常的1%,但4~14天仍能稳步恢复到正常,说明芥子气中毒后极少量的残存CFUs仍具有快速自我更新及增殖、分化的功能。以上结果提示,素称“拟放射性”毒剂的芥子气对造血组织的毒性作用与辐射损伤相比,存在着质与量的差别。
To understand the characteristics of haemato-toxicology of mustard gas, we observed the changes of CFUs and CFUc in bone marrow and compared them with ~ (60) Co irradiation. The dose-survival curve of CFUs under mustard gas is an exponential straight line with an early shoulder region characterized by an n of 5.6 and a D_0 value of 13.7 mg / kg whereas the exponential straight line of ~ (40) Co irradiation has n = 1.1, D_0 = 125rad. The survival rate of mustard gas CFUs under the same LD50 was 10 times higher than that of radiation, indicating that CFUs were sensitive to mustard but less sensitive than radiation. With 3 / 4LD_ (60) and other toxic doses of mustard gas, CFUs proliferation began early, 4 to 18 days after poisoning returned to normal, and ~ (60) Co irradiation after 7 to 36 days to return to normal half. The CFU or CFUc survival rate of the first day after 35 mg / kg high dose of mustard poisoning was lower than the normal 1%, but still steady returned to normal after 4 to 14 days, indicating that a very small amount of residual CFUs after mustard poisoning still has rapid self Update and proliferation, differentiation of the function. The above results suggest that there is a qualitative and quantitative difference between the toxic effects of mustard gas, which is called “pseudo-radioactive” poison, on hematopoietic tissue and radiation damage.
