目的观察新加良附方联合5-Fu对人胃癌细胞SGC-7901裸鼠移植瘤生长抑制作用及其对肿瘤组织中PTEN表达的影响。方法建立移植性人胃癌细胞SGC-7901裸鼠移植瘤模型,随机分为模型对照组、5-FU组及新加良附方高、中、低剂量组、联合给药组(新加良附中剂量+5-FU组)6组。连续给药10d,计算肿瘤抑制率,检测PTEN抑癌基因的表达。结果联合给药组抑瘤率为54%,与模型对照组比较,有统计学意义(P<0.05);与新加良附高剂量比较联合给药组抑瘤率升高明显,有统计学意义(P<0.05);联合给药组上调肿瘤组织PTEN蛋白表达,与模型对照组比较,有统计学意义(P<0.05);与新加良附高剂量组比较,联合给药组PTEN蛋白表达升高明显(P<0.05)。结论联合给药组抑瘤率高于新加良附高剂量组及5-FU组,在调节PTEN表达方面优于新加良附高剂量组及5-Fu组,显示了两药协同作用的优势。
OBJECTIVE To observe the inhibitory effect of Xinjialiangfuxian combined with 5-Fu on the growth of human gastric cancer cell SGC-7901 xenografted in nude mice and its effect on the expression of PTEN in tumor tissue. Methods The transplanted human gastric cancer cell SGC-7901 xenografted nude mice model was established and randomly divided into model control group, 5-FU group and Xinjialiangfuxian high, medium and low dose groups, combined with administration group Dose + 5-FU group) 6 groups.
