目的 探讨多粘菌素B(PMB)对内毒素 (LPS)刺激的巨噬细胞Toll4、TNF α、IL 6mRNA表达的抑制作用。方法 应用鲎试剂法测定PMB对LPS的中和作用 ,运用鼠腹腔巨噬细胞分离培养 ,并加入PMB与LPS培养 3h ,RT PCR检测Toll4、TNF α、IL 6mRNA转录水平变化。结果 PMB具有较强的中和LPS能力 ,其IC50 约为 4 .35± 0 .91μmol·L-1;PMB1.75mg·ml-1能够显著抑制 1μg的LPS刺激的鼠腹腔巨噬细胞Toll4、TNF α、IL 6mRNA的表达水平 (P <0 .0 5 )。结论 PMB可能通过对LPS的中和作用 ,进而抑制LPS刺激细胞因子的mRNA表达
Objective To investigate the inhibitory effect of polymyxin B (PMB) on the expression of Toll4, TNF α and IL 6 mRNA in macrophages stimulated by endotoxin (LPS). Methods The neutralization effect of PMB on LPS was determined by the method of Reagent. The cells were isolated and cultured with mouse peritoneal macrophages and cultured in PMB and LPS for 3 hours. The transcriptional levels of Toll4, TNFα and IL-6 mRNA were detected by RT-PCR. Results PMB had a strong capacity of neutralizing LPS and its IC50 was about 4.35 ± 0.91μmol·L-1. PMB1.75mg · ml-1 could significantly inhibit 1μg LPS-induced Toll-4, α, IL 6 mRNA expression levels (P <0.05). Conclusions PMB may inhibit the LPS-stimulated cytokine mRNA expression through the neutralization of LPS
