目的探讨APRIL(TNFSF13)/BLyS系统及其受体BCMA、TACI(TNFRSF13B)、BAFFR mRNA在原发性胆汁性肝硬化(PBC)患者外周血中的表达水平和临床意义。方法设计特异性引物和荧光探针,实时检测PCR产物的荧光强度,检测42例正常健康者和56例PBC患者外周血中APRIL/BLyS系统及其受体mRNA表达水平,采用全自动酶标仪测定PBC患者血清中细胞因子IL-2、IL-4、IL-10和TNF-α的水平。结果与健康者相比,PBC患者外周血中APRIL(TN-FSF13)、BLyS及其受体BCMA的mRNA表达均升高(P<0.01),APRIL、BLySmRNA表达与BCMAmRNA表达有显著相关性(分别为r=0.607和r=0.559,P均<0.01);APRILmRNA的表达与血清IL-2水平正相关(r=0.462,P<0.05),BLySmRNA的表达与血清IL-2、TNF-α水平正相关(r=0.740,P<0.01;r=0.381,P<0.05)。结论 APRIL/BLyS系统及其受体可能参与PBC的疾病进程,为监控PBC的病情和有效治疗提供了新的途径。
Objective To investigate the expression and clinical significance of APRIL (TNFSF13) / BLyS system and its receptors BCMA, TACI (TNFRSF13B) and BAFFR mRNA in peripheral blood of patients with primary biliary cirrhosis (PBC). Methods Specific primers and fluorescent probes were designed to detect the fluorescence intensity of PCR products in real time. The mRNA expression levels of APRIL / BLyS and its receptors in peripheral blood of 42 normal controls and 56 PBC patients were detected. Serum levels of IL-2, IL-4, IL-10 and TNF-α were measured in PBC patients. Results The mRNA expression of APRIL (TN-FSF13), BLyS and its receptor BCMA in peripheral blood of PBC patients was significantly higher than that of healthy controls (P <0.01), and APRIL and BLyS mRNA were significantly correlated with BCMA mRNA expression (R = 0.607, r = 0.559, P <0.01). The expression of APRIL mRNA was positively correlated with serum IL-2 level (r = 0.462, P <0.05). The expression of BLyS mRNA was positively correlated with the levels of IL-2 and TNF- (R = 0.740, P <0.01; r = 0.381, P <0.05). Conclusion The APRIL / BLyS system and its receptors may participate in the disease process of PBC, which provides a new way to monitor the condition and effective treatment of PBC.
