目的探讨pERK蛋白在胃癌及癌旁组织中的表达及其与肿瘤恶性行为的相关关系。方法收集98例胃癌患者的胃癌组织及癌旁组织标本,应用免疫组化检测其中pERK蛋白表达情况,并根据胃癌组织pERK蛋白表达阳性情况将其分为pERK蛋白(+)组74例、pERK蛋白(-)组24例。采用荧光定量PCR法检测胃癌组织中增殖、侵袭、自噬基因mRNA表达量,评估pERK蛋白阳性表达对上述基因mRNA表达量的影响。结果胃癌组织中pERK蛋白阳性表达率显著高于癌旁组织,差异有统计学意义(P<0.05)。pERK蛋白(+)组增殖基因EZH2、PIK3CD、I2PP2A mRNA表达量高于pERK蛋白(-)组,WTX mRNA表达量低于pERK蛋白(-)组;侵袭基因E-cadherin、Syndecan-1 mRNA表达量低于pERK蛋白(-)组,HPA-1 mRNA表达量高于pERK蛋白(-)组;自噬基因Beclin1、MAP1LC3 mRNA表达量低于pERK蛋白(-)组,差异有统计学意义(P<0.05)。结论 pERK蛋白在胃癌组织中阳性表达率高,且存在pERK蛋白阳性表达的胃癌组织肿瘤恶性程度高。
Objective To investigate the expression of pERK protein in gastric cancer and its adjacent tissues and its relationship with malignant behavior of tumor. Methods The gastric cancer tissues and adjacent normal tissues were collected from 98 gastric cancer patients. The expression of pERK protein was detected by immunohistochemistry. According to the positive expression of pERK protein in gastric cancer tissues, 74 cases were divided into pERK (+) group, 74 cases of pERK protein (-) group of 24 cases. The mRNA expression of proliferation, invasion and autophagy in gastric cancer tissues was detected by real-time quantitative PCR, and the effect of pERK protein expression on mRNA expression of these genes was evaluated. Results The positive expression rate of pERK protein in gastric cancer tissues was significantly higher than that in paracancerous tissues, the difference was statistically significant (P <0.05). The mRNA expression levels of EZH2, PIK3CD and I2PP2A in pERK (+) group were higher than that in pERK (-) group and WTX mRNA was lower than that in pERK (-) group. The expressions of E-cadherin and Syndecan-1 mRNA The expression of HPA-1 mRNA in pERK protein (-) group was lower than that in pERK protein (-) group, while the expression of Beclin1 and MAP1LC3 mRNA was lower than that in pERK protein (-) group (P < 0.05). Conclusion The positive expression rates of pERK protein in gastric cancer tissues are high, and the expression of pERK protein in gastric cancer tissues with high expression of malignancy is high.
