探讨了15和30天尾吊法模拟失重对大鼠肿瘤坏死因子-α(TNF-α)的含量及骨髓细胞对GM-CSF反应性的影响。采用放射免疫分析方法检测腹腔巨噬细胞在LPS刺激下产生TNF-α的含量,3H-TdR掺入法检测骨髓细胞对GM-CSF的反应性。结果表明,模拟失重15天后,TNF-α含量有下降的趋势,30天后TNF-α含量显著减少(P<0.05)。15天后,骨髓细胞对GM-CSF的反应性显著下降(P<0.01),30天后,两组间无明显改变。结果提示,15及30天模拟失重降低免疫功能,TNF-α含量的减少可能与骨髓细胞的增殖功能有关。
The effects of simulated weightlessness on the levels of tumor necrosis factor-α (TNF-α) and the response of bone marrow cells to GM-CSF in 15 and 30 days tail-suspended method were investigated. The level of TNF-α produced by LPS stimulated by peritoneal macrophages was detected by radioimmunoassay. 3H-TdR incorporation was used to detect the reactivity of bone marrow cells to GM-CSF. The results showed that after 15 days of simulated weight loss, the content of TNF-α tended to decrease, and the content of TNF-α decreased significantly after 30 days (P <0.05). After 15 days, the reactivity of myeloid cells to GM-CSF was significantly decreased (P <0.01). After 30 days, there was no significant change between the two groups. The results suggest that 15 and 30 days simulated weightlessness reduce immune function, TNF-αlevels may be related to the proliferation of bone marrow cells.
