正常人血清和淋巴组织浆细胞中各IgG亚类所占比例分别为IgG_1 60%~65%,IgG_2 25%~30%,IgG_3 5%~10%,IgG_4≤5%;而在IgG~+B细胞中的比例为IgG_1≈40%,IgG_2≈50%,IgG_3≈8%,IgG_4≤5%,看来在B细胞分化的不同阶段调控IgG_1和IgG_2亚类的表达有差异.用正常人PBMC分离纯化T,B细胞.纯化的B细胞加PWM培养,并于其中分别加入PWM诱导的T细胞培养上清或不同组合的重组IL-2,IL-4,IL-6和IFN-γ,以测定各类淋巴因子对B细胞产生IgG亚类的影响;加入PHA刺激的T细胞于自体B细胞中测定T-B直接接触时对IgG产生的影响.
