目的探讨温肾活血、益气升清的益筋方对实验性自身免疫性重症肌无力(MG)小鼠的治疗机理。方法采用重症肌无力患者的血清被动免疫小鼠,建立实验性自身免疫性重症肌无力模型。分组后分别给予益筋方、强的松灌胃21d,处死动物、采血。利用流式细胞技术,以T细胞及其亚群、IL-2为指标,观察益筋方、强的松对实验性重症肌无力小鼠细胞免疫和细胞因子的影响。结果强的松、益筋方均能升高CD3+,降低CD4+/CD8+比值;降低IL-2。结论益筋方能通过影响T细胞及其亚群、细胞因子,调控细胞免疫和体液免疫,起到治疗重症肌无力的作用。
Objective To investigate the therapeutic mechanism of “Yinhen Huoxue” and “Yiqi Shengqing” Yijin Fang on experimental autoimmune myasthenia gravis (MG) mice. Methods The serum of patients with myasthenia gravis was passively immunized to establish an experimental autoimmune myasthenia gravis model. After being grouped, Yijin Fang and prednisone were administered intragastrically for 21 days, animals were sacrificed, and blood was collected. Using flow cytometry, T cells and their subpopulations and IL-2 as indicators were used to observe the effects of Yijin Fang and prednisone on cellular immunity and cytokines in mice with experimental myasthenia gravis. Results Prednisone and Yijin can all increase CD3+, decrease the ratio of CD4+/CD8+ and decrease IL-2. Conclusion Yijin Fang can play a role in the treatment of myasthenia gravis by affecting T cells and their subpopulations, cytokines, regulating cellular immunity and humoral immunity.
