目的:观察粉防己碱(Tet)对雄黄中毒大鼠的排砷作用。方法:取大鼠60只,随机分为正常对照组、雄黄中毒模型组(模型组)、二巯基丙醇磺酸钠(DMPS)组、Tet高、中、低剂量组,每组10只。每组每日灌胃给予雄黄,各给药组分别给予不同剂量的药物腹腔注射,连续6周。给药期间,收集大鼠24 h尿以热消化-二乙氨基二硫代甲酸银(DDC-Ag)比色法的改良法测定尿砷含量,每周一次。停药后心脏取血用同样方法测定血砷含量。结果:各给药组大鼠24 h尿砷含量与模型组比较,Tet高、中剂量组以及低剂量组前4周的尿砷差异有极显著性(P均<0.01),低剂量组的第5、第6周尿砷量也有显著性差异(P<0.05);与DMPS组比较,仅低剂量组显示差异(P<0.05)。各给药组大鼠血砷含量与模型组比较,具有极显著性差异(P均 <0.01);与DMPS组比较,仅低剂量组有显著性差异(P<0.05)。结论:Tet对雄黄中毒大鼠具有明显的促进砷排泄作用。其作用与剂量呈正比例关系。
Objective: To observe the effect of tetrandrine (Tet) on arsenic in rats exposed to realgar. METHODS: A total of 60 rats were randomly divided into normal control group, realgar poisoning model group (model group), dimercaptopropionic acid sodium sulfonate (DMPS) group, Tet high, middle, and low dose groups, with 10 rats in each group. In each group, realgar was given by gavage daily, and each dose group was given different doses of the drug intraperitoneally for 6 consecutive weeks. During the dosing period, the 24-hour urine was collected and the arsenic content in urine was measured by a thermodigestion-diethylaminodithiocarbamate (DDC-Ag) colorimetric method. After the drug was stopped, blood was drawn from the heart and blood arsenic content was measured by the same method. RESULTS: Compared with the model group, the arsenic levels in 24 h urine of all groups were significantly higher than those in the model group (P<0.01). There was also a significant difference in urinary arsenic between the 5th and 6th week (P<0.05). Compared with the DMPS group, only the low dose group showed a difference (P<0.05). Compared with the model group, the arsenic content in the blood of each treatment group had a very significant difference (P<0.01); compared with the DMPS group, only the low dose group had a significant difference (P<0.05). CONCLUSION: Tet has a significant effect on promoting arsenic excretion in realgar-intoxicated rats. Its role is proportional to the dose.
