目的:研究芹菜素对间歇低氧后幼鼠部分脑区p38MAPK的影响。方法:SPF级健康雄性SD幼鼠(3~4周龄)40只,随机分为4组:间歇低氧组(IH组)、对照组(C组)、芹菜素+二甲基亚砜组(Q组)和二甲基亚砜组(R组)。建立间歇低氧幼鼠模型,以RT-PCR法和W esternb lot法分别测幼鼠海马及前额叶皮层p38MAPKmRNA和磷酸化p38MAPK(p-p38)蛋白的表达。结果:IH组幼鼠海马、前额叶皮层的p38MAPK mRNA及p-p38蛋白均明显高于C组(P均<0.01),Q组的表达则明显低于R组(P均<0.01)。结论:芹菜素可降低间歇低氧幼鼠脑区p38MAPK的激活,这可能对减轻间歇低氧后脑损伤具有积极意义。
Objective: To study the effect of apigenin on p38MAPK in partial brain regions of young rats after intermittent hypoxia. Methods: Forty SPF healthy male SD rats (3 to 4 weeks old) were randomly divided into 4 groups: intermittent hypoxia group (IH group), control group (C group), apigenin + dimethyl sulfoxide group (Q group) and dimethylsulfoxide group (R group). The models of intermittent hypoxic preconditioning were established. The expressions of p38MAPKmRNA and p-p38MAPK (p-p38) protein in the hippocampus and prefrontal cortex of young rats were detected by RT-PCR and Western blot respectively. Results: The expression of p38MAPK mRNA and p-p38 protein in hippocampus and prefrontal cortex in IH group were significantly higher than those in C group (P <0.01). The expression in Q group was significantly lower than that in R group (P <0.01). Conclusion: Apigenin can reduce the activation of p38MAPK in the brain of intermittent hypoxic rats, which may have a positive effect on reducing brain injury after intermittent hypoxia.
