目的:观察槟榔对离体大鼠结肠平滑肌收缩运动的影响,并初步探讨其机制。方法:制备大鼠结肠平滑肌肌条,以9g/LNaCl溶液(NS)、槟榔为对照组,分别记录单独使用槟榔和阻断剂(酚妥拉明、维拉帕米和阿托品)孵育后槟榔引起的收缩效应。结果:不同浓度槟榔对离体大鼠结肠平滑肌的促进作用明显,与NS对照组比较,槟榔0.1g/L、1g/L、10g/L浓度时引起的肌条收缩最大振幅和面积均有统计学意义(P<0.05);与单独槟榔组比较,阻断剂维拉帕米和阿托品对槟榔引起的肌条收缩有抑制作用,其最大收缩振幅和面积均有统计学意义(P<0.05);而阻断剂酚妥拉明对肌条的收缩振幅和面积的影响均无统计学意义(P>0.05)。结论:槟榔对大鼠结肠平滑肌的收缩活动有兴奋作用,且呈正相剂量关系,引起的收缩效应可被M受体阻断剂阿托品和Ca2+阻断剂维拉帕米阻断,而未能被α-受体阻断剂酚妥拉明阻断。
Objective: To observe the effect of areca nut on contraction of isolated rat colonic smooth muscle and to explore its mechanism. Methods: The colon smooth muscle strips of rats were prepared and betel cells were induced by betel nut and blockers (phentolamine, verapamil and atropine) after being incubated with 9g / LNaCl solution (NS) and betel nut respectively Contraction effect. Results: The effects of different concentrations of areca on the smooth muscle of isolated rat colonic muscle were obvious. Compared with the NS control group, the maximum amplitude and area of contractions induced by betel nut at 0.1 g / L, 1 g / L and 10 g / L were statistically (P <0.05). Compared with betel nut group alone, the antagonist verapamil and atropine could inhibit the contraction of muscle strips induced by betel nut, with the maximum amplitude and area of contraction being statistically significant (P <0.05) The effect of blocker phentolamine on contraction amplitude and area of muscle strips was not statistically significant (P> 0.05). CONCLUSION: Betel nut is excited by the contractile activity of rat colonic smooth muscle and has a positive phase dose relationship. The contractile effect can be blocked by the retinoic acid receptor blocker atropine and the verapamil, a Ca2 + antagonist, but not by Phentolamine, an α-receptor blocker, is blocked.
