目的 探讨胶质瘤细胞血小板源生长因子 (PDGF)BB表达水平与肿瘤间质血管(SBV)增生的关系。方法 用原位杂交、免疫组化和HE染色观察了 73例人胶质瘤。结果 73例中 ,仅 8例 ( 11% )有少许SBV表达PDGFBmRNA ,SBV的PDGFα受体、β受体和酪氨酸磷酸化蛋白 (P Tyr)的阳性率分别为 95 9%、 34 3%和 6 1 6 % ,四者间均有显著性差异 (P <0 0 0 5 )。 6 2例( 84 9% )的肿瘤细胞表达PDGFBmRNA ,其表达水平与SBV的密度及P -Tyr阳性率均呈平行递增 ,三者也均随肿瘤恶性等级升高而相应增加 ( P <0 0 5~ 0 0 0 1)。结论 本组结果提示胶质瘤细胞普遍表达PDGFBB ,后者能以旁分泌方式在刺激胶质瘤SBV增生方面发挥重要作用
Objective To investigate the relationship between the expression of platelet-derived growth factor (PDGF) BB and proliferation of tumor interstitial vessels (SBV) in glioma cells. Methods 73 cases of human gliomas were observed by in situ hybridization, immunohistochemistry and HE staining. Results Of the 73 patients, only 8 (11%) had SBV expressing PDGFB mRNA. The positive rates of PDGFα receptor, β receptor and tyrosine phosphorylation protein (P Tyr) in SBV were 95 9% and 34 3%, respectively. And 6 6%, there was a significant difference between the four (P <0 0 05). Sixty-two (84.9%) tumor cells expressed PDGFB mRNA, and their expression levels increased in parallel with the SBV density and P-Tyr positive rate. The three also increased with the increase of malignant grade of the tumor (P <0. 5 to 0 0 0 1). Conclusion The results of this study suggest that glioma cells generally express PDGFBB, which can play an important role in stimulating SBV proliferation in gliomas by paracrine mode.
