【目的】为研究一株筛选自大黑山林间土壤下的生防细菌,以形态学、16S rDNA进行鉴定。同时对其抑菌蛋白进行分离。【方法】采用对峙平板法进行初筛、复筛,用抑菌圈法测定抑菌效果,并镜检菌丝效果。通过形态学观察,生理生化及16S rDNA基因序列分析研究其分类。以SephacryI S-200分离抑菌蛋白。【结果】生防细菌K1对灰霉病具有稳定的抑菌效果,抑菌直径达到45 mm;16S rDNA鉴定为解淀粉芽孢杆菌。无菌发酵液通过SephacryI S-200分离得到一种抗菌蛋白。SDS-PAGE显示其分子量为55k Da。【结论】筛选出一株生防解淀粉芽孢杆菌K1,分离得到一种分子量为55k Da的抗菌蛋白。
【Aim】 To identify a biocontrol bacterium isolated from the soil of Daheishan Forest by morphological and 16S rDNA. At the same time its antimicrobial protein separation. 【Method】 The initial screening and re-screening were carried out by the counter-plate method, the antibacterial effect was determined by the inhibition zone method, and the mycelial effect was examined by microscopy. The morphological observation, physiological and biochemical and 16S rDNA gene sequence analysis of its classification. Antibacterial proteins were isolated using Sephacryl S-200. 【Result】 Biocontrol bacterium K1 had a stable antibacterial activity against Botrytis cinerea, with an antibacterial diameter of 45 mm. 16S rDNA was identified as Bacillus amyloliquefaciens. Anaerobic fermentation broth was separated by Sephacryl S-200 to obtain an antimicrobial protein. SDS-PAGE showed a molecular weight of 55 kDa. 【Conclusion】 A strain of Bacillus amyloliquefaciens K1 was screened out and an antimicrobial protein with a molecular weight of 55 kDa was isolated.
