丙戊酸钠是一种抗癫痫新药,为了能找到它的合适治疗剂量及药物代谢动力学研究,本文报导了快速测定血浆中微量丙戊酸钠的气液色谱分析法。将含有丙戊酸钠的血浆酸化后,用氯仿萃取其中游离出的丙戊酸,以每次1微升氯仿萃取液在涂渍了3%FFAP的Gas Chrom Q色谱柱上直接进样,以N_2为载气,氢火焰离子检测器检测,进行气液色谱分析。分析结果,丙戊酸的色谱峰面积与其相应浓度(在0.1~0.5微克/微升)为直线关系。本分析方法操作简便快速,定量准确,重现性好,能适用于临床。
Sodium valproate is an antiepileptic drug. In order to find a suitable therapeutic dose and pharmacokinetic study, we report the gas-liquid chromatographic method for the rapid determination of trace sodium valproate in plasma. After the plasma containing sodium valproate was acidified, the free valproic acid was extracted with chloroform and directly injected on a Gas Chrom Q column coated with 3% FFAP with 1 microliters each of chloroform extract to N_2 carrier gas, hydrogen flame ion detector detection, gas-liquid chromatography. Analysis results, valproic acid chromatographic peak area and its corresponding concentration (at 0.1 ~ 0.5 micrograms / microliters) is a linear relationship. The analysis method is simple, rapid, accurate, reproducible, and suitable for clinical application.
