目的建立高效液相色谱法测定刺五加中紫丁香苷和异秦皮啶的含量。方法采用KromasilC18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-体积分数为1%乙酸水溶液(B),梯度洗脱;流速为1.0 mL.min-1;检测波长为265 nm(0 min),341nm(11 min)。结果紫丁香苷和异秦皮啶质量浓度分别在26.0~435 mg.L-1(r=0.999 7)和0.270~4.50 mg.L-1(r=0.999 6)内与峰面积呈良好的线性关系;平均回收率分别为98.01%(RSD=1.2%,n=6)和97.53%(RSD=1.1%,n=6)。结论该方法可同时测定刺五加药材中紫丁香苷和异秦皮啶的含量。
Objective To establish a HPLC method for the determination of syringin and isofraxid in Acanthopanax senticosus. Methods The mobile phase consisted of acetonitrile (A) - 1% acetic acid in water (B) with gradient elution at a flow rate of 1.0 mL · min-1 on a Kromasil C18 column (250 mm × 4.6 mm, 5 μm) 265 nm (0 min), 341 nm (11 min). Results The syringin and isofenidone concentrations showed good linear relationship with the peak area in the range of 26.0-435 mg.L-1 (r = 0.999 7) and 0.270-4.50 mg.L-1 (r = 0.999 6) The average recoveries were 98.01% (RSD = 1.2%, n = 6) and 97.53% (RSD = 1.1%, n = 6), respectively. Conclusion This method can be used to simultaneously determine the content of syringin and isofraxid in Acanthopanax senticosus.
