目的:探讨血管紧张素Ⅱ(Ang Ⅱ)和表皮生长因子(EGF)是否经不同的蛋白激酶C(PKC)亚基介导而激活CCDPK产生促培养新生大鼠心肌成纤维细胞增殖。方法:Western Bloting和[~3H]thymidine参入法。结果:PDBU对Ang Ⅱ诱导的CCDPK活性和DNA合成速率无显著影响。而EGF诱导的CCDPK活性下降了65%,DNA合成速率下降了38%。PD 98059对Ang Ⅱ和EGF诱导的CCDPK活性分别下降了70%和72%。结论:PKC-ζ介导了Ang Ⅱ对CCDPK的激活及心肌成纤维细胞的增殖效应。
Objective: To investigate whether angiotensin Ⅱ (Ang Ⅱ) and epidermal growth factor (EGF) are mediated by different protein kinase C (PKC) subunits and activate CCDPK to promote the proliferation of cultured neonatal rat cardiac fibroblasts. Methods: Western Bloting and [~ 3H] thymidine incorporation. Results: PDBU had no significant effect on the activity of CCDPK induced by Ang Ⅱ and DNA synthesis rate. The EGF-induced CCDPK activity decreased by 65%, DNA synthesis rate decreased by 38%. PD 98059 decreased the CCDPK activity induced by Ang II and EGF by 70% and 72%, respectively. Conclusion: PKC-ζ mediates the activation of CCDPK by Ang Ⅱ and the proliferation of cardiac fibroblasts.
