目的探讨卵巢切除所致肾虚对Ⅱ型胶原诱导的小鼠关节炎(CIA)的影响及细胞因子的作用。方法于DBA/1小鼠皮下注射Ⅱ型胶原制作关节炎模型;以依赖株细胞增殖反应法测定IL-2、IL-6含量。结果卵巢切除可使关节炎小鼠脾单个核细胞的IL-2和IL-6含量显著增加;同时小鼠关节肿胀程度、滑膜组织增生、软骨损伤及软骨下骨的破坏均明显加重。结论IL-2、IL-6与CIA发病过程密切相关,卵巢切除所致肾虚可使CIA病变进一步加重,在一定程度上反映了此法所致肾虚与IL-2、IL-6之间的内在联系
Objective To investigate the effects of kidney deficiency induced by ovariectomy on type Ⅱ collagen induced arthritis (CIA) and the effects of cytokines. Methods The model of arthritis was established by subcutaneous injection of type Ⅱ collagen into DBA / 1 mice. IL-2 and IL-6 levels were determined by cell-dependent proliferation assay. Results Ovariectomy could significantly increase the levels of IL-2 and IL-6 in the spleen mononuclear cells of the arthritic mice. At the same time, the degree of joint swelling, synovial tissue hyperplasia, cartilage damage and subchondral bone destruction were significantly aggravated. Conclusions IL-2 and IL-6 are closely related to the pathogenesis of CIA. Kidney deficiency caused by ovariectomy may further aggravate CIA.
