目的探讨激活素A(Activin A)及激活素结合蛋白(Follistatin,FS)在急性酒精性肝损伤小鼠肝组织中的表达。方法通过1次/12 h给予小鼠5 g/Kg酒精连续灌胃3次,复制急性酒精性肝损伤小鼠模型,采用实时定量RT-PCR检测小鼠肝组织Activin A及FS mRNA表达水平,免疫组织化学染色观察小鼠肝组织Activin A及FS蛋白的表达水平。结果急性酒精性肝损伤小鼠肝组织Activin A mRNA和蛋白表达水平均显著高于对照组(P<0.01);FS mRNA和蛋白表达水平与对照组比较,差异均无统计学意义(P>0.05)。结论急性酒精性肝损伤小鼠肝组织Activin A-FS表达失衡,以Activin A升高为主,提示Activin A-FS系统失衡可能与酒精性肝损伤有关。
Objective To investigate the expression of activin A and follistatin (FS) in the liver of mice with acute alcoholic liver injury. Methods The mouse model of acute alcoholic liver injury was established by intragastric administration of 5 g / Kg alcohol to mice 3 times a day for 12 h. Real-time quantitative RT-PCR was used to detect the expression levels of Activin A and FS mRNA. Immunohistochemical staining was used to observe the expression of Activin A and FS protein in liver of mice. Results The levels of Activin A mRNA and protein in the liver of mice with acute alcoholic liver injury were significantly higher than those in the control group (P <0.01). There was no significant difference in the expression levels of FS mRNA and protein between the two groups (P> 0.05 ). Conclusion The expression of Activin A-FS in hepatic tissue of mice with acute alcoholic liver injury is unbalanced, which is mainly caused by the increase of Activin A, suggesting that the imbalance of Activin A-FS system may be related to alcoholic liver injury.
