本文采用制备大鼠肺条件培养基进行单层软琼脂法培养大鼠骨髓造血祖细胞(GM—CFU),进行定量研究。结果显示肺条件培养基的刺激因子(GM—CSF)活性好,15%浓度的CM刺激2.5月龄大鼠骨髓每4×10~5个有核细胞中形成细胞集落92.5±23.8。定量研究结果表明,老年大鼠的GM—CFU明显下降,如青年大鼠(2.5月龄)的GM—CFU为92.5±23.8,而老年大鼠(24月龄)的GM—CFU则为25.4±9.6。进一步观察发现有随年龄增长而下降的趋势,提示老化的机体其骨髓造血功能是低下的。
In this study, rat bone marrow hematopoietic progenitor cells (GM-CFU) were cultured in rat lung conditioned medium and subjected to single-layer soft agar for quantitative studies. The results showed that the lung conditioned medium stimulating factor (GM-CSF) activity is good, 15% concentration of CM stimulated 2.5-month-old rat bone marrow to form a cell colony of 92.5 ± 23.8 per 4 × 10 ~ 5 nucleated cells. Quantitative studies showed that GM-CFU in old rats decreased significantly. For example, GM-CFU in young rats (2.5 months old) was 92.5±23.8, whereas in old rats (24 months old), GM-CFU was 25.4± 9.6. Further observations revealed that there was a trend of decline with age, suggesting that the body of the aging bone marrow hematopoietic function is low.
