目的:研究模板纯化与否对HBV拉米呋啶耐药突变检测的检出率影响。方法:采用离心柱纯化法及直接煮沸裂解法分别处理HBV DNA定量小于104IU/ml的60例患者血清标本,用同一HBV拉米呋啶耐药突变检测荧光PCR试剂盒检测,比较其检出率。结果:HBV DNA定量小于104IU/ml的60例患者血清标本,离心柱纯化组和煮沸裂解组的检出率分别为68.3%和38.3%。结论:低病毒载量的HBV血清样品经离心柱纯化模板后可以明显提高HBV拉米呋啶耐药突变检测的检出率。
Objective: To study the effect of template purification on the detection rate of HBV lamivudine resistance mutation. Methods: Serum samples of 60 patients with less than 104 IU / ml of HBV DNA were respectively processed by centrifugal column purification and direct boiling lysis. The same HBV lamivudine resistance mutation detection fluorescent PCR kit was used to detect the detection rate . Results: The serum samples of 60 patients whose HBV DNA was less than 104 IU / ml were detected by the methods of centrifugation and boiling. The detection rates were 68.3% and 38.3% respectively. Conclusion: The detection rate of HBV lamivudine resistance mutations in HBV serum samples with low viral load purified by centrifugation column can be significantly improved.
