目的:观察天冰调督胶囊对印防己毒化学点燃癫痫模型大鼠学习记忆行为学改变及海马组织NMDAR1表达的影响。方法:SD大鼠腹腔注射印防己毒素(1.5 mg/kg)连续30 d,制作反复发作癫痫模型,点燃模型大鼠随机分为模型组、天冰调督胶囊低(0.4 g/kg)、高(0.8 g/kg)剂量组、丙戊酸钠组及拉莫三嗪组,采用Morris水迷宫检测各组大鼠学习记忆行为学,RT-PCR检测海马组织NMDAR1 mRNA,Western-blot和免疫组化法检测NMDAR1蛋白表达情况。结果:治疗60 d后,与模型组比较,天冰调督胶囊低、高剂量组大鼠水迷宫实验寻找平台时间显著缩短(P<0.05或P<0.01),通过平台次数显著增加(P<0.01);NMDAR1 mRNA含量均增加(P<0.05或P<0.01);在CA3、DG区NMDAR1表达均增强(P<0.05)。结论:天冰调督胶囊可能通过调节NMDAR1水平改善癫痫学习记忆等认知障碍。
Objective: To observe the effect of Tian Bing Tiao Du capsule on learning and memory abilities and hippocampal NMDAR1 expression in rats with epilepsy model. Methods: Sprague-Dawley rats were intraperitoneally injected with picrotoxin (1.5 mg / kg) for 30 days to make a recurrent seizure model. The rats were randomly divided into model group and Tianbingdu capsule (0.4 g / kg) (0.8 g / kg), sodium valproate and lamotrigine groups. Morris water maze test was used to evaluate the learning and memory behaviors of hippocampus. NMDAR1 mRNA, Western-blot and immunohistochemistry were detected by RT-PCR. Method to detect the expression of NMDAR1 protein. Results: After 60 days of treatment, compared with the model group, Tianbingchu capsule lowers and high doses rats water maze test to find the platform time was significantly shorter (P <0.05 or P <0.01), the number of platforms significantly increased (P < (P <0.05 or P <0.01). The expression of NMDAR1 in CA3 and DG were increased (P <0.05). Conclusion: Tianbingdusu capsule may improve cognition disorder such as epilepsy learning and memory by regulating NMDAR1 level.
