茶碱(THP)有兴奋中枢神经、扩张支气管等作用,用于心脏疾患、支气管哮喘、新生儿窒息等症的治疗,作者以比较简单的方法处理成批样品,用薄层层析(TLC)光密度扫描同时测定血清中THP、CAF,结果良好。[试剂]①标准液:THP、CAF各为10μg/ml的水溶液。②内标准液(I.S.):配成7-(2-羟乙基)-茶碱100μg/ml水溶液。③抽提溶剂:氯仿:异丙醇=19:1(v/v)。④0.1N盐酸。⑤TLC展开溶剂:氯仿:丙酮:异丙醇:氨=6:4:2:0.5。[仪器]①TLC板:メルケ制TLC板(硅胶60F_(254),5×10cm)。②微量加样器(10μl)。③展开槽:ャトロェ,11×3×8cm。④双波长层析
Theophylline (THP) stimulates the central nervous system and expands the bronchus. It is used for the treatment of heart disease, bronchial asthma, and neonatal asphyxia. The authors processed batches of samples in a relatively simple manner and used thin-layer chromatography (TLC). Simultaneous determination of THP and CAF in serum by densitometry showed good results. [Reagents] 1 standard solution: Each of THP and CAF was an aqueous solution of 10 μg/ml. 2 internal standard solution (I.S.): A 7-(2-hydroxyethyl)-theophylline 100 μg/ml aqueous solution was prepared. 3 Extraction solvent: chloroform: isopropyl alcohol = 19:1 (v/v). 40.1N hydrochloric acid. 5TLC development solvent: chloroform: acetone: isopropanol: ammonia = 6:4:2:0.5.
