目的:建立桃红清血胶囊中2,3,5,4′-四羟基二苯乙烯-2-O-β-D葡萄糖苷含量的HPLC测定方法,为工业生产的质量控制提供依据。方法:采用高效液相色谱法(HPLC)测定桃红清血胶囊中2,3,5,4′-四羟基二苯乙烯-2-O-β-D葡萄糖苷的含量,具体条件为:Shim-packVP-ODSC18柱(150.0mm×4.6mm,5μm),流动相为乙腈-水(17∶83),检测波长为322nm,柱温为30℃。结果:2,3,5,4′-四羟基二苯乙烯-2-O-β-D葡萄糖苷在0.284~2.272μg范围内有良好线性关系,回归方程为Y=-25678+1204880X,r=0.9999;平均回收率99.4%,(RSD)为2.29%(n=6);以精密度实验、稳定性实验和重复性实验考察的RSD分别为0.46%、2.89%和2.39%。经测试并结合生产实际情况限定桃红清血胶囊中2,3,5,4′-四羟基二苯乙烯-2-O-β-D葡萄糖苷含量不少于0.8mg/粒。结论:HPLC检测桃红清血胶囊中2,3,5,4′-四羟基二苯乙烯-2-O-β-D葡萄糖苷含量的方法准确、简单可行,重复性好,可作为桃红清血胶囊质量控制方法。
Objective: To establish an HPLC method for the determination of 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside in Taohongqingxue Capsule and provide the basis for the quality control of industrial production. Methods: The contents of 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside in Taohongqingxue Capsule were determined by high performance liquid chromatography (HPLC). The specific conditions were Shim- PackVP-ODSC18 column (150.0 mm × 4.6 mm, 5 μm) with acetonitrile-water (17:83) as the mobile phase at a detection wavelength of 322 nm and a column temperature of 30 ° C. Results: There was a good linear relationship between 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucosidase in the range of 0.284 ~ 2.272μg, the regression equation was Y = -25678 + 1204880X, r = 0.9999. The average recovery was 99.4% (RSD) 2.29% (n = 6). The RSDs were 0.46%, 2.89% and 2.39%, respectively, according to the precision test, stability test and repeatability test. Tested and combined with the actual production of limited peach red clear blood capsule 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside content of not less than 0.8mg / tablets.
