Ginsenosides are the principal components responsible for the pharmacological and biological activities of ginseng.Following oral administration of Panax ginseng extract,the ginsenosides and its metabolites were identified and quantified in rat plasma,urine and faeces by LC-MS/MS.Rat plasma,urine and faeces samples were collected from 0 to 24 h after the oral administration of Panax ginseng extract(1000mg/kg).In the metabolic study,ginsenosides and its deglycosylated metabolites were characterized by comparing the retention time,accurate mass,and characteristic MS/MS fragment ions with standard compounds.The ginsenosides excreted in urine and faeces were ginsenosides Rg1,Re,Rf,Rb1,Rc,Rb2,Rd,Rg2,Rh1,F1,F2,Rg3,PPT,compound K,and Rh2 were found,and only compound K was detected in plasma samples collected after oral administration.This is the first time quantitative information on the metabolites of ginsenosides in rat plasma,urine and faeces after oral administration of Panax ginseng extract has been reported.These results will help to understand the pharmacokinetics and metabolism of ginsenosides,a principle pharmacologically active compound of ginseng.
