目的研究肺炎链球菌RNA对细胞分泌细胞因子的影响。方法提取肺炎链球菌总RNA,分别用不同浓度的RNA作用于肺腺癌细胞A549和巨噬细胞U937,同时以LPS作为对照作用于两细胞,观察细胞分泌细胞因子情况。结果细菌RNA刺激细胞后,其分泌IL-12的能力显著提高且呈剂量依赖关系,当RNA浓度为40μg/ml时,IL-12分泌量提高5倍。而细胞分泌IL-18、TNF-α、IFN-γ的能力与RNA的作用量无直接关系。结论肺炎链球菌RNA具有诱导细胞分泌细胞因子IL-12的能力,是重要的病原相关的分子模式。
Objective To study the effect of Streptococcus pneumoniae on cytokines secreted by cells. Methods The total RNA of Streptococcus pneumoniae was extracted. The lung adenocarcinoma A549 cells and the macrophage U937 cells were treated with different concentrations of RNA respectively. Meanwhile, LPS was used as a control to treat both cells, and cytokine secretion was observed. Results The ability of bacterial RNA to stimulate IL-12 secretion was significantly increased in a dose-dependent manner. When the concentration of RNA was 40μg / ml, IL-12 secretion increased by 5 times. However, the ability of cells to secrete IL-18, TNF-α and IFN-γ has no direct relationship with the amount of RNA. Conclusions Streptococcus pneumoniae RNA has the ability of inducing cells to secrete cytokine IL-12 and is an important pathogen-related molecular pattern.
