目的观察牛磺酸对脓毒症大鼠肺损伤的保护作用,并探讨其作用机制。方法 24只雄性SD大鼠分为3组:假手术组、模型组和牛磺酸组。盲肠结扎并穿孔法建立脓毒症大鼠动物模型,模型组及牛磺酸组分别于术后0,6,12 h,尾静脉注射生理盐水5 m L·kg~(-1)、牛磺酸200 mg·kg~(-1),假手术组只开腹不手术,不给药。各组于术后24 h处死大鼠,留取标本。HE染色观察各组肺组织形态学改变,用末端标记法测定细胞凋亡指数,以蛋白免疫印迹法测定各组肺组织中人X盒结合蛋白1(XBP-1)、活化的转录因子4(ATF-4)及C/EBP同源蛋白(CHOP)表达。结果与模型组相比,牛磺酸组病理改变不明显,牛磺酸组凋亡指数(0.26±0.03)%明显低于模型组(0.27±0.03)(P<0.05),而肺组织XBP-1、ATF-4、CHOP蛋白的表达均明显弱于模型组(P<0.05)。结论牛磺酸对脓毒症大鼠肺损伤有保护作用,其机制与减轻内质网应激介导的细胞凋亡相关。
Objective To observe the protective effect of taurine on lung injury in septic rats and to explore its mechanism. Methods Twenty-four male SD rats were divided into 3 groups: sham operation group, model group and taurine group. Cecal ligation and perforation to establish animal model of sepsis in model group and taurine group at 0, 6, 12 h after the operation, the tail vein injection of saline 5 m L · kg -1, Acid 200 mg · kg ~ (-1), sham operation group only laparotomy without medication. Rats in each group were killed at 24 h after operation and samples were taken. The morphological changes of lung tissue were observed by HE staining. The apoptotic index was determined by end-labeling method. The expression of XBP-1, activated transcription factor 4 ATF-4) and C / EBP homologous protein (CHOP) expression. Results Compared with the model group, the pathological changes of taurine group were not obvious. The apoptosis index (0.26 ± 0.03)% in taurine group was significantly lower than that in model group (0.27 ± 0.03) (P <0.05) 1, ATF-4, CHOP protein expression were significantly weaker than the model group (P <0.05). Conclusion Taurine has a protective effect on lung injury induced by sepsis in rats. The mechanism is related to the reduction of endoplasmic reticulum stress-induced apoptosis.
