为观察硫酸庆大霉素对培养的小鼠耳蜗螺旋神经节细胞(SGC)的细胞生存数和细胞突起长度的影响,在单离培养的出生1~5d的NIH小鼠SGC培养液中,加入不同浓度的硫酸庆大霉素(25,50,100mg/L)。培养14d后,标本用4%多聚甲醛固定,甲苯胺蓝染色,在光镜下计数SGC生存数,目镜测微尺下测量细胞突起长度。结果显示,实验组与对照组间、不同浓度实验组间的细胞生存数及细胞突起长度无明显差异(P>0.05)。提示硫酸庆大霉素对小鼠耳蜗SGC的生存和生长无明显影响,氨基糖甙类抗生素对SGC的影响可能是继发性的。
To observe the effects of gentamicin sulfate on cell viability and cell protrusion length of cultured mouse spiral ganglion cells (SGC), NIH mouse SGC culture medium 1 to 5 days old Gentamycin sulfate at different concentrations (25, 50, 100 mg / L). After cultured for 14 days, the specimens were fixed with 4% paraformaldehyde and stained with toluidine blue. The number of surviving SGCs was counted under a light microscope and the length of the cells was measured under an eyepiece micrometer. The results showed that there was no significant difference in cell survival number and cell protrusion length between the experimental group and the control group (P> 0.05). It is suggested that gentamycin sulfate has no significant effect on the survival and growth of mouse cochlear SGC. The effect of aminoglycoside antibiotics on SGC may be secondary.
