目的研究系统性红斑狼疮(SLE)患者外周血中CD4+CD25+T细胞亚群的比率改变。方法采用微量全血三色标记法,以流式细胞技术检测SLE患者外周血CD4+、CD4+CD25+、CD4+CD25+CD45RO+T细胞亚群,分析阳性细胞百分率和平均荧光强度(Meanfluorescenceintensity,MFI)。结果SLE患者外周血中CD4+T细胞百分率,活动期(29.29%±8.86)%和非活动期(29.55±8.96)%均低于正常对照(37.41±3.29)%(P均<0.05);CD4+CD25+T细胞百分率SLE活动期(10.30±5.60)%显著高于正常对照(5.39±1.43)%(P<0.05),非活动性SLE病人组(5.63±2.49)%和正常组差异无显著性(P>0.05);CD4+CD25+CD45RO+T细胞百分率SLE活动期(3.96±3.51)%显著高于正常对照(1.39±0.63)%,非活动期(0.75±0.66)%显著低于正常对照(P均<0.05)。平均荧光强度分析表明SLE患者组和正常组的CD25和CD45RO抗原密度差异并无显著性(P均>0.05)。结论SLE患者外周血中存在CD4+CD25+及CD4+CD25+CD45RO+T细胞亚群比率异常增高,且与病情活动性相关。
Objective To investigate the ratio of CD4 + CD25 + T cell subsets in peripheral blood of patients with systemic lupus erythematosus (SLE). Methods CD4 +, CD4 + CD25 +, CD4 + CD25 + CD45RO + T cell subsets in peripheral blood of SLE patients were detected by flow cytometry. The percentage of positive cells and mean fluorescence intensity (MFI) . Results The percentages of CD4 + T cells in peripheral blood of SLE patients (29.29% ± 8.86%) and inactive period (29.55 ± 8.96%) were lower than those of normal controls (37.41 ± 3.29%) (P <0.05) + CD25 + T cells in active SLE group (10.30 ± 5.60)% was significantly higher than that in normal control group (5.39 ± 1.43)% (P <0.05), but there was no significant difference between inactive SLE group and normal group (5.63 ± 2.49)% (P <0.05). The percentage of CD4 + CD25 + CD45RO + T cells in SLE group was significantly higher than that in normal control group (3.96 ± 3.51)% vs 0.75 ± 0.66% Control (P <0.05). Mean fluorescence intensity analysis showed no significant difference in CD25 and CD45RO antigen density between SLE patients and normal controls (all P> 0.05). Conclusion There is an abnormally high ratio of CD4 + CD25 + and CD4 + CD25 + CD45RO + T cells in the peripheral blood of SLE patients, which is related to the activity of the disease.
