目的 探讨神经营养因子诱导成年大鼠骨髓基质细胞成神经细胞的可能性,为神经再生及视神经疾病的治疗提供一种新方法。方法 从大鼠骨髓中分离培养骨髓基质细胞并传至第6代,诱导前24h光用含1μg/L bFGF及 20%FBS的DMEM培养液培养,以促迸细胞分裂,然后用神经生长因子(NGF)和脑源性神经生长因子(BDNF)作诱导剂,观察细胞形态的变化,并采用免疫组织化学法检测诱导后的细胞表达Nestin、NF、NSE、Map-2、GFAP等特异性标志物。结果 诱导后1h,部分细胞的形态已发生改变,5h后大部分细胞不仅在形态上表现为神经元样特征,而且Nestin、NF、NSE及Map-2等特异性抗体呈阳性表达,而GFAP抗体表达呈阴性。结论 神经营养因子能将骨髓基质细胞诱导成神经元样细胞。
Objective To explore the possibility of neurotrophic factor-induced adult rat bone marrow stromal cells into neurons and to provide a new method for the treatment of nerve regeneration and optic nerve diseases. Methods Bone marrow stromal cells were isolated and cultured from rat bone marrow and passaged to passage 6. The cells were cultured in DMEM medium containing 1μg / L bFGF and 20% FBS for 24h before induction to promote cell division, and then stimulated with nerve growth factor ( NGF and BDNF were used as inducers to observe the changes of cell morphology. Immunohistochemistry was used to detect the expression of Nestin, NF, NSE, Map-2, GFAP and other specific markers . Results The morphological changes of some cells were observed 1 h after induction. Most of the cells showed neuron-like features after 5 h, and the specific antibodies such as Nestin, NF, NSE and Map-2 were positive. GFAP antibody Negative expression. Conclusion Neurotrophic factors can induce bone marrow stromal cells into neuron-like cells.
