1990年4月,辽宁省本溪市虹鳟鱼种场的虹鳟稚鱼爆发大规模疾病,患鱼长约2.5cm,体重约0.2克,以体侧肌肉线状出血为典型特征,死亡率近100%.取病鱼组织处理后接种于冷水性鱼细胞,出现明显的细胞病变(CPE).按照Reed Muench两氏法在96孔板的CHSE细胞中,测定细胞培养物的病毒滴度,最高达到10~(6.3)T CID50/0.1ml.感染病毒的细胞用1%营养琼脂糖履盖后可形成0.5~1.5mm的蚀斑.经理化特性和生物学特性测定表明,分离出的病毒对氯仿敏感,不耐热、不耐酸,在细胞内复制的最适
In April 1990, a large-scale disease broke out in juvenile rainbow trout at the rainbow trout farm in Benxi City, Liaoning Province. The fish suffered from a length of about 2.5 cm and a weight of about 0.2 grams. The typical muscle haemorrhages of the body side were characterized by a mortality rate of nearly 100% The diseased fish tissues were inoculated into cold-water fish cells and showed obvious cytopathic effect (CPE) .The virus titer of the cell culture was determined according to the method of Reed Muench in 96-well CHSE cells up to 10 ~ (6.3) T CID50 / 0.1ml.The virus-infected cells could form 0.5-1.5mm plaques with 1% nutrient agarose.The physicochemical properties and biological properties showed that the isolated virus was sensitive to chloroform , Heat-labile, acid-labile, optimal for intracellular replication
