采用高效液相色谱法(HPLC),在硅胶涂敷纤维素三-(4-甲基苯甲酸酯)手性固定相上拆分非洛地平。研究了异丙醇体积含量、流速及柱温对非洛地平对映体拆分行为的影响。同时还模拟了在制备分离条件时,增大进样体积和进样浓度对非洛地平拆分的影响。并结合色谱理论和热力学理论对非洛地平的手性识别机理进行探讨。实验表明在正己烷∶异丙醇=90∶10,流速0.8mL/m in,柱温30℃时,非洛地平对映体可实现基线分离。
Felodipine was resolved on a silica gel coated cellulose tri- (4-methylbenzoate) chiral stationary phase using high performance liquid chromatography (HPLC). The effects of isopropanol volume fraction, flow rate and column temperature on the enantiomeric resolution of felodipine were investigated. At the same time, we also simulated the effect of increasing the injection volume and injection concentration on the felodipine resolution when preparing the separation conditions. The chiral recognition mechanism of felodipine was discussed based on chromatographic theory and thermodynamic theory. Experiments show that in n-hexane: isopropyl alcohol = 90:10, the flow rate of 0.8mL / min, the column temperature of 30 ° C, felodipine enantiomer baseline separation can be achieved.
