应用尿激酶原(proUK)基因重组复制缺陷型腺病毒载体,防治家兔髂动脉内膜剥脱后新生内膜的增生。结果显示:导入proUK基因三周后,血管组织内proUK基因的mR-NA及其表达产物明显高于导入野生型腺病毒对照组;血管造影及切片显示血管狭窄程度较轻,新生内膜与中膜面积比(I/M)明显低于对照组。提示:血管内膜剥脱后导入proUK基因,通过对抗血管内膜损伤后的血栓形成和血小板的沉积,从而抑制由血栓及血小板促发的新生内膜过度增生,具有潜在的应用价值。
Recombinant urokinase (proUK) gene was used to replicate defective adenovirus vector to prevent neointimal hyperplasia after intima dissection in rabbit iliac artery. The results showed that after three weeks of introduction of proUK gene, the mR-NA of proUK gene and its expression product in vascular tissue were significantly higher than that of the wild-type adenovirus control group. Angiography and section showed that the degree of vascular stenosis was mild. Neointimal and middle Membrane area ratio (I / M) was significantly lower than the control group. It is suggested that the introduction of proUK gene after endometrial stripping has potential application value by inhibiting the thrombosis and platelet deposition after intimal injury and thus inhibiting the neointimal hyperplasia induced by thrombus and platelet.
