肿瘤坏死因子受体(TNFR)是肿瘤坏死因子(TNF)发挥其生物学作用的关键性物质。利用聚合酶链反应(PCR)技术,扩增并改造55KDTNFRcDNA,成功地扩增出编码可溶性肿瘤坏死因子受体成熟肽的序列,并在该序列的5’端和3’端分别引入EcoRI和HindⅢ识别序列,使之便于与载体重组,为获得亚克隆及在大肠杆菌高效表达奠定基础。
Tumor necrosis factor receptor (TNFR) is a key substance that tumor necrosis factor (TNF) plays its biological role. The polymerase chain reaction (PCR) technique was used to amplify and transform 55KDTNFR cDNA to successfully amplify the mature peptide encoding soluble Tumor Necrosis Factor Receptor (TNFR). At the 5 ’end and 3’ end of the sequence, EcoRI and HindⅢ Identification of the sequence, making it easy to recombine with the vector, in order to obtain subclones and efficient expression in E. coli.
