目的:使用一氧化氮合酶(NOS)抑制模型评价罗布麻叶不同成分群的活性。方法:通过检测样品对NOS在催化L-精氨酸(L-Arg)氧化分解L-CIT和一氧化氮(NO)过程中同步引起还原型辅酶Ⅱ(NADPH)氧化为(NADP)+而产生荧光值变化的影响,间接反映罗布麻叶成分群的抑制的NOS活性。结果:首次发现罗布麻叶中成分群Ⅱ(黄酮类成分)具有抑制NOS的活性,IC50=0.501mg·mL-1。结论:罗布麻叶中的黄酮类成分具有潜在的保护脑组织损伤的作用。
OBJECTIVE: To evaluate the activity of different components of Apocynum leaf using a nitric oxide synthase (NOS) inhibition model. METHODS: Samples were tested for the simultaneous oxidation of NOS by oxidation of reduced L-arginine (L-Arg) to L-CIT and nitric oxide (NO) to cause the oxidation of reduced coenzyme II (NADPH) to (NADP)+. The effect of changes in fluorescence values indirectly reflects the inhibitory NOS activity of Apocynaceae leaf populations. Results: It was found for the first time that component II (flavonoids) in A. venetum leaves inhibited NOS activity with IC50=0.501 mg·mL-1. Conclusion: Flavonoids in Apocynum leaf have the potential to protect brain tissue from injury.
