目的建立杜仲及相关制剂中松脂醇二葡萄糖苷(PDG)的含量测定方法,测定不同来源的杜仲及杜仲制剂的PDG含量。方法高效液相色谱法测定杜仲药材中PDG的含量,色谱柱为Agilent Eclipse XDB-C18(150×4.6mm,0.46μm),流动相:甲醇-水(26:74),流速:1.0mL·min-1,检测波长:230nm,检测温度为25℃,进样量:10μL。结果PDG在0.05080~0.5080mg·mL-1范围内呈良好线性(r=0.9999);炮制后的杜仲中PDG含量均高于0.105%,较生药材要高;6种杜仲制剂中均没有检测出PDG。结论本方法简便、快速、准确性高、重复性好,适用于杜仲及其相关制剂中PDG的含量测定。
Objective To establish a method for determination of pinoresinol diglucoside (PDG) in Eucommia ulmoides and related preparations, and determine PDG content of Eucommia ulmoides Oliv. and Eucommia ulmoides preparations from different sources. Methods The content of PDG in Eucommia ulmoides Oliv. was determined by HPLC. The chromatographic column was Agilent Eclipse XDB-C18(150×4.6mm, 0.46μm). The mobile phase was methanol-water(26:74). The flow rate was 1.0 mL·min. -1, detection wavelength: 230 nm, detection temperature 25°C, injection volume: 10 μL. Results The PDG was linear in the range of 0.05080-0.5080 mg·mL-1 (r=0.9999). The content of PDG in processed Eucommia ulmoides Oliv. were higher than 0.105%, higher than that of raw herbs. None of the 6 Eucommia ulmoides preparations were detected. PDG. Conclusion The method is simple, rapid, accurate and reproducible. It is suitable for the determination of PDG in Eucommia ulmoides and related preparations.
