海岛棉“7124”是选育得到的高抗黄萎病的棉花品种。在黄萎病菌诱导的7124幼苗中,经硫酸铵盐析和电泳分离,获得了一种热不稳定的蛋白质。体外平板实验,该蛋白质在100mM柠檬酸-柠檬酸钠缓冲液中,低于1μg即能明显抑制黄萎病菌孢子萌发和生长。利用显微摄影技术观察到了液体环境中病菌孢子的萌发抑制现象。上述抗菌蛋白质的发现、分离和纯化将为棉花抗黄萎病基因的分离提供基础。
Island cotton “7124” is a bred cotton variety with high resistance to Verticillium wilt. In the 7124 seedlings induced by Verticillium dahliae, a thermolabile protein was obtained by ammonium sulfate salting-out and electrophoresis separation. In vitro plate experiments, the protein in 100mM citrate - sodium citrate buffer, less than 1μg that can significantly inhibit Verticillium wilt spore germination and growth. The inhibition of the spore germination of the bacteria in the liquid environment was observed by using the photomicrography technique. The discovery, isolation and purification of the above antimicrobial proteins will provide the foundation for the isolation of cotton genes related to Verticillium dahliae.
