目的:证实博来霉素A5抑制血管生成和抗肿瘤转移作用。方法:将含有0.5μg博来霉素A5的载体置于胚龄7d的鸡胚绒毛尿囊膜(CAM)上,48h后观察抑制血管生成情况。Lewis肺癌皮下接种C57BL6小鼠6d后,在移植瘤周围及其基底部多处局部注射博来霉素A5,每鼠每次0.6mg,每5天1次,共3次,接种后25d处死小鼠,观察皮下移植瘤血管生成抑制情况和对肺转移的抑制作用。结果:博来霉素A5可使载体周围CAM新生血管明显减少甚至消失。Lewis肺癌皮下移植瘤体积小,颜色苍白,毛细血管稀少,很少出血,坏死常见。肺转移抑制率为80%。结论:博来霉素A5能强烈抑制鸡胚CAM和Lewis肺癌皮下移植瘤的血管生成,瘤周局注具有抗转移作用。提示博来霉素A5是一有潜力的血管生长抑制剂。
OBJECTIVE: To confirm the effect of bleomycin A5 on angiogenesis and anti-tumor metastasis. Methods: The vector containing 0.5 μg bleomycin A5 was placed on chick embryo chorioallantoic membrane (CAM) for 7 days and observed for 48 hours to inhibit angiogenesis. Lewis lung cancer was inoculated subcutaneously with C57BL6 mice for 6 days. Bleomycin A5 was injected locally at several sites around the xenograft and its basal ganglia. Each rat received 0.6 mg once every 5 days for 3 times. The mice were sacrificed 25 days after inoculation Rats were observed subcutaneous xenograft angiogenesis inhibition and inhibition of lung metastasis. Results: Bleomycin A5 significantly decreased or even disappeared the CAM neovascularization around the carrier. Lewis lung cancer subcutaneous tumor size is small, pale, capillaries sparse, rarely bleeding, necrosis common. Pulmonary metastasis inhibition rate was 80%. Conclusion: Bleomycin A5 strongly inhibits the angiogenesis of subcutaneously transplanted tumor in CAM and Lewis lung carcinomas, and has the anti-metastasis effect of peritumoral injection. Prompted Bleomycin A5 is a potential angiogenesis inhibitor.
