目的研究大鼠不同发育阶段脑髓鞘化的发展及规律。方法应用LFB髓鞘染色、MBP免疫组化及Wes-tern blot观察不同发育阶段大鼠脑髓鞘的分布及变化。结果LFB及MBP免疫组化染色E18、P0、P2均未见着色,P7大鼠胼胝体中部少量LFB着色,P15两种染色均呈阳性,P30着色加深,与P90、P720类似。通过Western blot检测MBP蛋白在孕中后期已有表达,出生后随日龄增长逐渐增加,至老年期表达量最高。结论大鼠脑髓鞘化从P15已经开始,P30接近成熟;观察髓鞘的最佳时间在30d以后。
Objective To study the development and regularity of myelination in different developmental stages of rats. Methods The distribution and changes of myelin sheath in different developmental stages were observed by myelin staining, MBP immunohistochemistry and Wes-tern blot. Results Immunohistochemical staining of LFB and MBP showed no staining in E18, P0 and P2. A small amount of LFB staining was found in the middle part of the corpus callosum of P7 rats. Both of P15 staining and P30 staining were similar to P90 and P720. The expression of MBP protein was detected by Western blot in the second trimester of pregnancy. After birth, the expression of MBP protein gradually increased with the increase of the age, and reached the highest in old age. Conclusion Myelinization in rat brain has started from P15 and P30 is almost mature. The best time to observe myelin after 30 days.
