目的:研究Eag1钾通道在前列腺癌(PCa)组织中表达的状况,为探讨其与PCa发生、发展机制的相关性及能否作为PCa的诊治靶点提供理论依据。方法:采用RT-PCR和免疫组化的方法,检测Eag1钾通道mR-NA和蛋白在雄激素依赖性前列腺癌(ADPCa)组织、雄激素非依赖性前列腺癌(AIPCa)组织和癌旁非瘤组织中的表达水平。结果:Eag1钾通道mRNA在ADPCa癌旁非瘤组织(A)、AIPCa癌旁非瘤组织(B)、ADPCa组织(C)和AIPCa组织(D)中表达相对系数分别为0.265±0.413、0.167±0.511、2.673±2.988和2.815±2.901,与相应非瘤组织相比,Eag1钾通道mRNA在前列腺癌组织中表达显著增高(P<0.05);Eag1钾通道蛋白在A、B、C和D组中阳性表达率分别为7.4%、6.7%、88.9%和86.7%,与相应非瘤组织相比,Eag1钾通道蛋白在癌组织中表达显著增高(P<0.05)。结论:Eag1钾通道可能参与了PCa的发生和进展的病理生理学过程,有望成为PCa诊治中有价值的靶点。
OBJECTIVE: To study the expression of Eag1 potassium channels in prostate cancer (PCa) tissues and to provide a theoretical basis for exploring its correlation with the occurrence and development of PCa and its potential as a diagnostic target for PCa. Methods: The expression of mR-NA of Eag1 potassium channel and its expression in the tissues of androgen-dependent prostate cancer (ADPCa), androgen-independent prostate cancer (AIPCa) and adjacent non-cancerous tissues were detected by RT-PCR and immunohistochemistry The level of expression in the tissue. Results: The relative expression coefficients of Eag1 potassium channel mRNA in ADCCa non-cancerous tissues (A), AIPCa non-cancerous tissues (B), ADPCa tissues (C) and AIPCa tissues (D) were 0.265 ± 0.413 and 0.167 ± 0.511,2.673 ± 2.988 and 2.815 ± 2.901, respectively. Compared with the corresponding non-neoplastic tissues, the expression of Eag1 potassium channel protein in cancer tissues was significantly increased (P <0.05). Conclusion: Eag1 potassium channels may play a role in the pathogenesis of PCa and its pathophysiological process, which is expected to be a valuable target in the diagnosis and treatment of PCa.
