目的:为了合成金属螯合四肽并制备其单克隆抗体。方法:从初始原料起,采用液相接肽等方法,合成三乙撑四胺钴(Ⅲ)-N-(α-羧基-苯乙基)-β-Ala-Phe-β-Ala-GlyOH半抗原,并将其偶联到载体蛋白BSA上制成抗原,免疫小鼠,采用杂交瘤方法制备单克隆抗体;结果:成功地合成了该金属螯合四肽,获得了8株单克隆抗体杂交瘤细胞系,并对这8株McAb类与亚类等特性进行了鉴定,结论:可根据需要化学合成一些分子量很小的肽,并制备其单克隆抗体。
Aim: To synthesize metal-chelated tetrapeptides and prepare their monoclonal antibodies. Methods: Starting from the starting materials, the liquid phase peptide and other methods were used to synthesize the cobalt (Ⅲ) -N- (α-carboxy-phenethyl) -β-Ala-Phe-β-Ala-GlyOH Antigens were prepared and conjugated to the carrier protein BSA to make antigens. The mice were immunized and the monoclonal antibodies were prepared by the hybridoma method.
