目的 研究气管内应用GM CSF重组腺病毒 (AdGM CSF)对实验性肺转移癌的治疗作用。方法 (1)小鼠气管内滴入AdGM CSF ,ELISA法测定局部及外周血GM CSF及相关因子的水平。 (2 )C5 7BL/ 6小鼠尾静脉注射Lewis肺癌 3LL细胞建立肺转移癌模型 ,观察气管内应用AdGM CSF对实验性肺转移癌的治疗作用 ,包括肺转移结节、动物存活期、生存率变化等 ,并比较NK细胞和CTL活性的不同。结果 (1)气管内应用AdGM CSF后 72h肺灌洗液中检测到GM CSF且高于外周血水平 ,对照组肺灌洗液及外周血中均未检测到。(2 )气管内滴入AdGM CSF对实验性肺转移癌有治疗作用 ,可使肺转移结节减少 ,动物的生存期延长及存活率升高 ,同时对NK细胞、CTL活性有增强作用。结论 气管内应用GM CSF重组腺病毒可有效表达并对实验性肺转移癌有治疗作用
Objective To study the therapeutic effects of intratracheal administration of GM CSF recombinant adenovirus (AdGM CSF) on experimental lung metastases. Methods (1) AdGM CSF was instilled into the trachea of mice. ELISA was used to determine the levels of GM CSF and related factors in local and peripheral blood. (2) C57BL/6 mouse tail vein injection of Lewis lung cancer 3LL cells was used to establish a lung metastatic carcinoma model. The effect of intratracheal administration of AdGM CSF on experimental metastatic lung cancer was observed, including lung metastasis nodule, animal survival, and survival rate. Change, etc., and compare the difference in NK cell and CTL activity. Results (1) GM CSF was detected in lung lavage fluid 72 h after administration of AdGM CSF and was higher than peripheral blood levels. No lung lavage fluid and peripheral blood were detected in the control group. (2) Intratracheal instillation of AdGM CSF has therapeutic effects on experimental lung metastases, which can reduce the number of metastatic nodules in the lungs, prolong the survival of animals, increase the survival rate, and increase the activity of NK cells and CTL. Conclusion Intratracheal application of GM CSF recombinant adenovirus can effectively express and treat experimental lung metastases
